Effects of osmotic gradients on vacuolar malic Acid storage: a basic principle in oscillatory behavior of crassulacean Acid metabolism.

چکیده
Malate synthesis by CO(2) dark fixation and malate accumulation in the vacuoles of leaf slices of Kalanchoë daigremontiana Hamet et Perrier, a plant performing crassulacean acid metabolism, occurs only in external solutions where the osmotic pressure difference between the cells and the medium is low. Conversely, malate loss from the vacuoles depends on a high osmotic pressure difference between the cells and the medium and is observed in media of low osmotic pressure. This suggests that the diurnal oscillations of malate levels in crassulacean acid metabolism leaf cells are regulated by osmotic gradients. These findings support a model which is introduced to explain how the rhythm of crassulacean acid metabolism may function in the intact plant.

A malic Acid permease in isolated vacuoles of a crassulacean Acid metabolism plant.
Vacuoles isolated from mesophyll protoplasts of the Crassulacean acid metabolism plant Bryophyllum daigremontianum take up [(14)C]l-malic acid from the medium. The initial rates of uptake show saturation kinetics (apparent K(m) about 1 millimolar). In competition experiments, the uptake rates of l-malic acid are greatly reduced in the presence of a surplus of d-malic and tartronic acid. A numbe...
متن کاملEffect of malic acid on bioactive components and antioxidant properties of sliced button mushroom (Agaricus bisporus) during storage
Background: White button mushroom (Agaricus bisporus) is one of the most sensitive agricultural crops after harvesting and its antioxidant properties tend to decrease during post-harvest storage with the passage of time. OBJECTIVES: The possible role of exogenous application of malic acid as dipping treatment on bioactive components and antioxidant properties of sliced white button mushroom dur...

The concentration of glucan, mucilage, soluble carbohydrates, and malic acid were determined in Opuntia bigelovii Engelm. during a 23-week period. The experiment began during the dry summer by irrigation to stimulate Crassulacean acid metabolism and was followed by 13 weeks of drought. After the 13-week drought period, the plants were irrigated throughout a 10-week period until late December. T...
متن کاملStudies in the Metabolism of Crassulacean Plants: The Diurnal Variation in Organic Acid and Starch Content of Bryophyllum calycinum.
Crassulacean plants in general undergo diurnal variation in the acidity of the leaf tissue. They share this behavior in greater or less degree with plants of a number of other families, but crassulacean plants are the classical material for demonstration of the phenomenon; Bryophyllum calycinum, in particular, has been repeatedly investigated. Most studies have dealt with the titratable acidity...

The mechanisms and the controlling factors of malate oxidation by mitochondria from leaves of Kalanchoë blossfeldiana Poelln. plants performing Crassulacean acid metabolism were investigated using Percollpurified mitochondria. The effects of pH and of various cofactors (ATP, NAD(+), coenzyme A) on malate dehydrogenase (EC 1.1.1.37) and malic enzyme (EC 1.1.1.39) solubilized from these mitochond...
